Jonathon Stevenson's dramatic late winner stunned the visitors, rock bottom of the Conference table.
RMI took the lead through Chris Simms, who drilled a low shot home after 14 minutes.
Matthew Langston, Glen Fuff and Robbie Simpson missed chances for Cambridge before Dave Sadler restored parity with a powerful strike from 18 yards.
And Stevenson then drilled in the winner deep into injury time.
